Prostate-specific antigen, or PSA, tests are used to detect semen in evidence. Nebraska State Patrol Crime Lab.

PSA tests are used to determine if a stain found at a crime scene, on a victim's clothing etc contains semen. The stain is rubbed with a cotton swab and what the swab picks up is dissolved in a liquid. This liquid is ten introduced to the PSA test. It is a quick way of determining if there is indeed semen / sperm in a particular stain, piece of evidence, on clothing etc.
